My lab is devoted to investigate fundamental mechanisms of transcriptional control underlying cellular function. We utilize human pluripotent stem cells to model development and complex diseases as well as innovative high throughput functional genomic and genetic tools (e.g. high throughput screening of functional non-coding regulatory sequences utilizing CRISPR tools, mapping the 3D genomic structure, mapping regulatory sequences by epigenomic profiling) to investigate how regulatory elements affect gene expression. Specifically, we focus on elucidating the causal relationship between genetic and epigenetic variations at regulatory sequences, e.g. enhancers and neurodevelopment and neurological diseases, and how these factors interplay to control gene regulation in mammalian cells.
